The rapifleX is by far the most advanced TOF/TOF system today, and was re-designed from the ground up with focus on speed, robustness and confidence to meet today’s highest demands for in-depth intact and top-down protein characterization, and high-performance, high-throughput mass spectrometry imaging (MSI). The rapifleX TOF/TOF is well-suited for detailed protein characterization in life science research and biopharmaceutical laboratories. The new rapifleX MALDI PharmaPulse solution is the latest addition to Bruker’s portfolio of innovative products designed to assist pharmaceutical, biotech and CRO customers to accelerate drug discovery. Based on the new 10 kHz laser rapiflex™ MALDI-TOF system, and incorporating an integrated automation solution, this is the first mass-spectrometry based, label-free uHTS (ultra-high throughput screening) solution, using standard 1,536 microtitre plates, or even 6,144 plates. The rapifleX MALDI PharmaPulse solution analyses a 1,536 MTP in < 8 minutes, and due to its extreme robustness it is suitable for primary screens of millions of compounds, with significantly lower cost per sample. Its label-free detection of substrates and products accelerates assay development, reduces complexity of screening, and potentially minimizes artifacts from interferences with the labels used in traditional uHTS technologies.

最新生物制药应用资讯 - the new DisulfideDetect workflow

The tertiary structure of therapeutic proteins is key to their activity and stability. Due to the complexity of these proteins and the fact that they will contain multiple disulfide bonds, analysis is a challenge – often requiring several LC/MS runs with the tryptic digests from reduced and non-reduced protein and a manual comparison of these two analyses.
The DisulfideDetect workflow allows the fully automated analysis of disulfide bonds in biopharmaceuticals, based on a single digest of the unreduced protein and without prior knowledge of enzyme specificity or native disulfide bonds. The workflow provides a semi-quantitative scrambling analysis as well.
