Vitamin D (where D refers to D2 and D3) is a fat soluble vitamin. The major physiological function of vitamin D metabolites is to maintain calcium and phosphate homeostasis. Vitamin D insufficiency can result in rickets in children and osteoporosis in adults. Vitamin D status has been associated with a variety of diseases including cancer, diabetes, cardiovascular disease, osteoporosis and multiple sclerosis. 25-Hydroxy vitamin D is the metabolite measured in blood serum to determine the vitamin D status of patients.
According to current industry standards, vitamin D deficiency is defined below:
Severe Deficiency: Hydroxyvitamin D < 6 ng/ml
Deficiency: Hydroxyvitamin D < 16 ng/ml
Insufficiency: Hydroxyvitamin D < 20 ng/ml
Normal: Hydroxyvitamin D > 20 ng/ml
The goal of this work was to develop a UHPLC/MS method with field free APCI source for measurement of hydroxyvitamin D in serum with LOQ ≤ 4 ng/ml
方法
HPLC: PerkinElmer Flexar FX-10
Column: PerkinElmer Brownlee HRes C18, 1.9 µm, 2.1 x 50 mm
Gradient: 77 % MeOH/H2O, 5 mM NH4COOH for 1 min, followed by linear gradient to 90 % MeOH/H2O, 5 mM NH4COOH in 2min, hold at 90 % MeOH/H2O, 5 mM NH4COOH for 2 min at flow rate of 0.5 ml/min.
MS: PerkinElmer Flexar SQ 300 MS with PerkinElmer field free APCI source
APCI Source Temperature: 300℃
Corona Current: 5 µA
