从入门到精通,SFC于医药工业之应用

 

超临界流体色谱技术 (Supercritical fluid chromatography, SFC) ,顾名思义,即流动相既不是气体也不是液体,而是使用超过临界温度和临界压力下的单一态流体(Supercritical Fluid)作流动相进行分离分析、制备纯化的色谱技术。

SFC的原理与液相色谱的原理类似,然而利用超临界流体(通常为二氧化碳CO2)取代有机溶剂或水作为主要流动相,从本质上来说是一种高效的正相色谱技术。作为流动相所使用的超临界流体CO2具有独特的低粘度和高扩散性能而成为一种智能溶剂,因具有与液体相近的密度,对各种物质有较强的溶解性,难挥发性和热稳定性差的物质分析均适用;同时,超临界流体的粘度近于气体,比液体约低1~2个数量级,而它的扩散系数在气体和液体之间,因而具有较快的传质速度和较小的传质阻力,分离过程中峰形变窄、柱效增加、分析速度加快。由此可见,SFCHPLCGC之长、避GCHPLC之短,具有独特的优越性,应用前景极为广阔。

由于SFC采用天然绿色超临界流体CO2作为溶剂,无需有机溶剂回收处理,省事经济环保,节能降耗又可持续使用,因而SFC生物医药、精细化工、生态环境、农业食品等行业具有独特的吸引力。

SFC在制药行业,尤其是手性药物开发领域,由于其能够有效地利用液相色谱中常用的手性固定相 (chiral stationary phaseCSP) 且与GCHPLC体系中使用的检测器完全兼容, SFC是一种理想的手性分离方法。因为它运行速度更快,溶剂用量更少,样品后处理更简单,总体上来讲,SFC是一种更经济、更绿色的方法,在手性化合物分离方面性能卓越,大有超越高效液相色谱法(HPLC)之趋势,为分离纯化和手性制备分离的有益替代方案。
