血球计数板计数误差的来源及解决方案—自动化计数

引言

血球计数板一直是实验室细胞计数的金牌标准。自从18世纪在法国第一次被用于分析病人的血液样本,血球计数板在过去几百年中已经得到一系列的重大发展,相比以前计数更为精确、使用更为简单,并最终形成了今天我们使用的样子。现在血球计数板计数仍然是所有细胞学研究的一个组成部分,然而其计数存在的问题由于自身固有的设计和使用方法并没有随着时间而消失。我们在这里将要列出造成血球计数板计数误差的来源, 并将讨论自动化计数是如何消除这些问题的。

血球计数板计数误差的来源

1. 人工失误(混匀、加样、稀释、计算错误以及人工操作误差)

a. 在对5个操作者的观察中,操作错误和随机错误分别占3.12%和7.8%[3]

b. James M. Ramsey做了一项实验,衡量取样区和稀释系数如何影响计数的准确性。

他测试了3个取样区面积(18, 9和4 mm2)及两个稀释系数(1:100 和 1:25)。取样面积减小时CVs值是升高的,稀释倍数的升高会降低CVs[4]

 c. Bane 发现,当同一个操作者去对同样的两份精液样品计数时,计数结果的差异55%归因于取样和移液问题, 45%归因于计数室和细胞计数问题 [5]。 Freund和Carol 展开的另外一项实验表明,不同操作者之间的计数差异能高达52%,而同一个操作者的计数差异为20%[5]

2. 多次计数以保证结果准确性的必要性

a. 1907年, John C. DaCosta 声明,为了得到精确的计数结果,很有必要取血液样品中的多滴血液分别进行计数[1].

b. Nielsen, Smyth和Greenfield得出结论, 为了得到10%, 15%和 20%的血球计数板计数准确性, ,必需的样品数分别为7份, 3份和2份,每份样品中分别包含180个, 200个和125个细胞[6]

c. 1881年, Lyon 和Thoma推测血球计数板的标准误差为 ,其中n即计数的细胞数目;

d. 1907年, William Sealy以“学生”的名义发布了他计数酿啤酒师的酵母的工作,他特地通过实验和数学模型计算了计数误差,公式也为[7,8]

3. 细胞均匀分布的要求

a. 1912年, James C. Todd将细胞分布不均匀列为计数误差的问题来源[1]

b. 学生也说有两项主要的计数误差来源,一为吸取的酵母样品不能够代表原液的浓度,另一个是随机取样时细胞在计数区域分布不均匀[7,8]

c. 1947年, 一篇文章提到血球计数板中的细胞浓度分布不均匀问题。最初的结果显示,离进样口最近和最远区域的浓度分别比平均浓度低3.5%和高3.5%[9]

4. 仪器及材料差异(栅格,深度,盖玻片,缓冲液类型以及移液器)

a. 结果显示计数室的计数误差和移液器(CV%)造成的计数误差分别在大约4.6% 和4.7%[10]

b. 在一项5个计数人员的计数实验中,移液器和血细胞计数器造成的误差分别为9.46% 和4.26%[3]

c. 1961年, Sanders和Skerry得出结论,盖玻片的位置能造成7.6%的计数差异[11]

d. 在关于不同稀释步骤的计数实验中,随着稀释步骤的增加,变异系数升高,每个血细胞计数系统的误差如下: Bürker-Türk (BT) (7.7%-12%), Thoma (6.6%-14.1%), Makler (19.8%-23.6%)[12]

解决血球计数板的计数问题

随着新技术的发展,如计算机技术、自动化软件、光学镜片、荧光染料、精密制造,以及现代技术如荧光显微技术、流式细胞术、图像细胞术,自动化已经解决了血球计数板存在的许多问题[13-25]

自动化计数解决:

人工操作误差 - 为了解决这个问题,自动化和机器人技术能够代替人工的样品操作和计数操作。.

加样误差 - 取样区越多、计数细胞越多,随机误差越小,但是需要时间越多。通过应用自动取样或者成像技术,成千上百万的细胞能在很短时间内被分析,提高了效率,并把分析中的随机误差降到最低。

移液和稀释误差 - 这些取决于操作者的操作经验。通过采用自动加样器或者自动液流系统,这个误差可以被降到最低[26]

材料误差 - 计数室的误差是由于不同品牌的血球计数板或者同一品牌不同批次间的差异造成的。这也可以通过自动细胞计数仪(细胞计数仪的选择,请查阅http://dakewe.com/product/view/id-71.html或百度文库http://wenku.baidu.com/view/f13faf4916fc700abb68fc54.html 中《细胞计数仪的选择》一文)增加取样量及减小随机误差来解决。

细胞分布不均匀 – 血球计数板不合适的清洗,或者盖玻片放置不正确将会产生误差。这些可以通过不使用计数室的细胞计数仪来消除,比如流式细胞仪。但是细胞样品中若是存在细胞团,基于液流计数的仪器将很难计数,而使用图像计数仪,细胞团可以使用图像分析算法计数聚集的细胞,这样可以提高细胞计数的准确性。

综述

血球计数板几百年来在生物医学研究中一直都是一个必备的工具,并且经历了很多的改进形成了今天研究者们使用的样子,然而它仍然会造成很多不可避免的计数误差。今天,现代化的自动细胞计数仪的使用已经很大程度上消除了许多出现误差的来源,提高了细胞计数的准确性和效率。
