酶联免疫吸附实验（简称ELISA）是继免疫荧光和放射免疫技术之后发展起来的一种免疫酶技术。此项技术自70年代初问世以来，发展十分迅速，目前已被广泛用于生物学和医学科学的许多领域。

ELISA是以免疫学反应为基础，将抗原、抗体的特异性反应与酶对底物的高效催化作用相结合起来的一种敏感性很高的试验技术。由于抗原、抗体的反应在一种固相载体—聚苯乙烯微量滴定板的孔中进行，每加入一种试剂孵育后，可通过洗涤除去多余的游离反应物，从而保证试验结果的特异性与稳定性。在实际应用中，通过不同的设计，具体的方法步骤可有多种。即：用于检测抗体的间接法，用于检测抗原的双抗体夹心法以及用于检测小分子抗原或半抗原竞争法等等。比较常用的是ELISA双抗体夹心法及ELISA间接法。

       ELISA载体主要以微量滴定板最为常用，专用于ELISA的产品称为ELISA板，国际上标准的微量滴定板为8x12的96孔式，反应孔容积约400微升。反应样品和试剂体积为100-200微升。为便于作少量标本的检测，有制成8联孔条或12联孔条的，放入座架后，大小与标准ELISA相同。ELISA板的特点是可以同时进行大量标本的检测，并可在特制的比色计上迅速读出结果。现在已有多种自动化仪器用于微量滴定板型的ELISA检测，包括加样、洗涤、保温、比色等多步骤，对操作的标准化极为有利。

 

    96孔板条

    为适应基因、蛋白、药物的研究发展而产生的高效率测试需求；为了减少试剂用量，降低成本，Microtier-Plate分析技术朝体积小，单位元密度高的方向来发展，以至于现在的基本孔数已经进展到了384或是1536个，每个反应孔缩小到100微升或20微升的容积，因而反应试剂的体积可以减少到40微升或10微升就足够完成一次测试了。目前为止，384孔板和1536孔板均为固定的，每次实验要求样品数量多（384个或1536个），因此不能随意使用。

    384孔酶标反应的最大优势在于其高通量和微型化，主要用于药物筛选和基因组学及蛋白组学的研究工作，既能进行大规模的研究以产生大量的实验数据，也能节省大量的研究试剂及研究样品。但是在常规的酶联免疫实验工作中，实验样品数不易凑足，整块的384孔板得不到推广使用。

这种“易拆分使用的384孔板”分为无色透明的微孔条用于普通酶联免疫吸附实验，黑颜色的微孔条用于化学发光酶联免疫反应实验和白颜色的微孔条用于荧光免疫反应实验。

       这种易拆分使用的384孔板由框架和反应孔条组成，每条反应孔由16个易掰开反应孔，24条反应孔条镶嵌到框架中即成384孔板，用户可以根据待测标本数量我多少随意拆开组合使用。

Enzyme-Linked Immunosorbent Assay (ELISA) is a immunoenzymatic technique developed after the invention of immunofluorescence technique and radioimmunology technique.

 

ELISA is a high sensitivity assay technique, based on the principles of immune reactions that combine specialized immune reaction and enzyme’s high catalyzing efficiency.

The immune reaction is happened on a solid-phase, the polystyrene micro plates, so after every time a reagent is loaded and incubated, the rest reactant can be washed away, as the result, the specificity and stability are guaranteed. In the real application, there are several different methods which are indirect ELISA used to determination antibody, paired-antibodies sandwich ELISA used to determination antigen and antigen competitive ELISA used to determination the low-molecular-weight antigen or hapten.

 

The most frequently used carrier of ELISA is microtiter-plates. Some microtiter-plates specifically used in ELISA are called ELISA plates. The international standard micro titer-plates are composed of 8 lines, 12 arrays, 96 wells. Every wells has a volume of 400 μl. The samples and reagents’ volume are often about 100-200 μl. There is 8/12 wells strip invented to make it easy to determine small-amount sample. These strips are fit with the ELISA plate frame. just as ELISA plates. The feature of ELISA plates is that you can determinate large-amount samples and you can read out the results directly by the colorimeter. Now, there have been many kinds of auto-instruments, which can sample, wash, and heat insulate and colorimetric, used to read ELISA plates. It’s much beneficial to the operational standardization.

To meet the demands of high efficient assay in gene, protein and drug research, to reduce the reagent and cost, micro titer-plate’s wells are made more and more smaller and denser, so, nowadays, the basic well-amount is 384/1536, every well’s capacity is reduced to 100/20μl. Thus, the reagent can be reduced to 40/10μl to finish one determination. Till now, all the 384/1536 microplates is in one piece, so it can only be used to determinate large-amount samples to avoid wasting.

A detachable 384 wells microplate.

The 384 wells ELISA plates have a great advantage in high-throughput and miniaturization application, so they are mainly used in drug screening, genomics and proteomic research, which are large-scale research and produce large amount of experimental data.However, in ordinary ELISA, not all 384 wells are used, so 384 wells microplates have not been popularized.

GenoMintel’s “detachable 384 wells microplates” solved this problem. No matter how much sample you have, you can use this kind microplates to run your ELISA. Because of the low capacity of the wells, compare with the 96 wells microplates, you can save 75% of your sample and reagent in the condition of having a same result. This “detachable 384 wells microplates” are divided into transparent strips used to run ordinary ELISA, black strips used to run chemiluminescence enzyme immunoassay and white strips used to run fluerescence immunoassy.

Type JR2008-A 96/384 wells auto microplate-washer was a new generation multifunction microplate-washer pushed out by GenoMintel in 2008. It is controled by microcomputer and can wash 96/384 wells microplates automatically.
