过表达

基因研究中常需要通过上调靶基因的表达来观察表型变化。过表达（Over-Expression，OE）是上调基因表达最常用的方法，其基本原理是将目的基因的编码区（coding sequence，CDS）构建到质粒或病毒载体中，导入细胞内使基因的表达量增加。根据用途，载体主要分为克隆载体、原核表达载体、真核表达载体、慢病毒载体、腺病毒载体、腺相关病毒载体、 逆转录病毒载体等。

过表达如何选择合适的载体呢？

首先我们需要考虑这几个方面:

1. 工具载体的表达时间与实验观察时间是否相符;

2. 工具载体可以容纳的片段大小与目的基因大小是否契合;

3. 工具载体对目的细胞、在体组织的转导是否足够高效。
