Bio-Rad S3e型荧光激活细胞分类仪

　　随着越来越多的研究人员在实验中加入了细胞分选，流式细胞仪的实验室就变得拥挤不堪。尽管免疫学家一直是这项技术的主要用户，但在细胞分选仪的预约名单上，其他领域的研究人员也日渐增多，这使得仪器的预约使用近乎饱和。有人打趣说，用上一台细胞分选仪，就像在闹市区找个停车位那么难。

　　然而，尽管有些用户需要高度复杂的分选，来获得特定的细胞群，但对于半数以上的用户，他们的分选需求要简单得多。一些分子生物学家刚刚进入这一领域，他们只是希望通过1-2个荧光蛋白或标记抗体来进行分选。根据Bio-Rad的调查，许多客户只是对特定细胞类型的特定蛋白感兴趣，或是希望分离表达GFP的所有细胞，仅此而已。对于他们而言，简单易用的分选仪可能是个更理想的选择。

　　Bio-Rad的S3细胞分选仪就是为满足这一群体的分选需求而设计的。它是一台高度自动化的系统，zei多能检测四种颜色，且体积小巧，大小仅为70 x 65 x 65 cm（宽x深x高），可放置在实验台上。这台仪器内置了液流和温控装置，因此无需其他的台面空间。

　　对于传统的细胞分选仪，技术人员往往需要几个月的培训，才能熟练地开展操作。但小型的实验室可能没有专门的人员来负责这个，或由于人员流动而出现断层。然而，有了S3细胞分选仪，这就完全不成问题。它的细胞分选操作相对简单，不需要专门的技术人员来完成。之前，土耳其的一个研究小组需要开展细胞分选，但他们没有任何经验。而在使用S3后仅一个星期，他们就能自由地操作。

　　分选原理

　　S3采用的是经典的jet-in-air（在空气中激发）的技术进行分选的。为了从多元化的细胞群体中获得感兴趣的细胞，研究人员首先要利用生物标志物标记细胞。然后让单细胞悬液通过流动柱，用激光照射，并将其分割成微小的液滴，每个都包裹着一个细胞。根据荧光信号，液滴在经过偏转板之前被带上不同的电荷。当液滴经过偏转板时，在高压电场的作用下偏转，落入各自的收集容器中。此过程每秒重复数万次，实现大量细胞的快速、高效分选。

　　自动化

　　S3细胞分选仪的简约大多源自其新颖的液滴延迟计算技术。为了获得zei佳的分选纯度，人们需要准确地确定液滴延迟，即细胞检测点与断点处之间的时间间隔。这一过程费时费力。而ProDrop™技术自动确定液滴延迟，并准确地计算出液滴延迟值，使得出错的机会更少。

　　据Bio-Rad细胞生物学业务部门的经理Steve Kulisch介绍，他们之前问过实验室技术人员，希望细胞分选的哪些过程有改变，得到的答案是：不必一直坐在那里，调整液滴延迟。这种痛苦使得他们重新考虑了细胞分选仪的设计。而自动化的液滴延迟大概就是来自于那个设计过程的zei大创新。

　　S3包含1或2个激光器及zei多4个荧光检测器和2个散射光检测器。它还有着用户可更换的滤光片设计，以便优化滤光片和荧光染料的组合。市场上多种滤光片都能与S3兼容。仪器标配了488 nm 100 mW二极管泵浦固态激光器。荧光素（FITC）、绿色荧光蛋白（GFP）、藻红蛋白（PE）、碘化丙啶（PI）以及其他常用的荧光染料都可用此激发并检测。选配的561 nm 100 mW固态激光器适合红色荧光蛋白家族及其他荧光染料。

　　当然，S3细胞分选仪并不打算取代BD或贝克曼家那些复杂的仪器。对于从事干细胞研究或肿瘤鉴定的研究人员，只需要四色分选，甚至单色GFP分选，这样一台简单易用的分选仪已经足够了。同时，它也让那些更为复杂的系统从简单应用中解放出来，真正应用到高度多重的免疫学分析中。这样，也能提高整个实验室的效率，帮助研究人员更好地安排他们的实验。

　　据Kulisch介绍，国外有个中心实验室就在自助服务区安装了S3细胞分选仪。研究人员可以预约他们的分选，如果是新手的话，需要接受30分钟的培训，然后开展分选，而几乎不需要监督。这几乎就像使用定量PCR仪那么简单。

　　细胞分选过程的简化，不仅能提高实验室的效率，也有望实现新的应用。Kulisch说：“随着人们更深入了解这种技术，他们更有可能提出这样的问题，‘细胞分选是否会带来不同？’。也许这个问题会带来遗传过程的发现，或让人了解到一个细胞群体比另一个更容易受到治疗的影响。这可能有助于人们更好地了解疾病状态。”
