人类干细胞来源的心肌细胞是制药公司和科研机构在研究中应用得比较多的样本。由于人类干细胞来源的心肌细胞具有和原代人类心肌细胞一样的遗传背景,它们的药理学特性应该非常相似,因此结合人类干细胞来源的心肌细胞与MED64多电极胞外记录系统的平台已成为了研究人类遗传性心肌疾病模型和药物筛选的主流。
只需要将hSC-CMs培养在MED记录电极上,就可以从MED64系统中记录和提取出基本的电生理参数,比如搏动频率、以场电位时程等。另外,由于电极的非侵入特性,稳定的记录可以持续非常长的时间,因此慢性药物的效果也能够在MED64系统上进行测量。不仅如此,MED64还可以使用任意电极输出刺激来Pace hSC-CMs,因此可用来研究频率依赖或使用依赖的药物作用。
