细胞外泌体/微囊泡解析专题(三)

B、D图: 显示两组样本外泌体CD47表达异常,乳腺癌组CD47明显表达减少,统计学差异P值=0.004说明巨噬细胞启动吞噬效力。
E图:在B、D图个选取N=60人份血液标本。 未配对t检验,P值<0.05.
F图:通过ELISA 测量健康人N=40人份,乳腺癌N=50份,CD47表达量,未配对t 检验,P值<0.01.

实验证明:超纳米级流式无法匹敌的散射光和分辨率可以检测不同细胞领域,通过健康人与乳腺癌患者CD47抗体表达量的不同,可以判定癌症疗效和预后指标。

微囊泡检测

日前,加拿大科学家Paproski R J等人首次研究证实,通过使用超灵敏纳米流式对胞外囊泡EVs的释放量进行绝对计数和鉴定(如图),发现配合纳米液滴,使用超声刺激可以显著增强肿瘤细胞EVs的释放,可使肿瘤细胞释放足够的EVs进入血液当中。同时,在这些胞外囊泡EVs中,Paproski R J和其同事成功检测到肿瘤相关蛋白、DNA、mRNA和miRNA。该研究成果为肿瘤的无创检测分析和遗传表型分析提供了重大的诊疗价值。
