Abstract Gives a procedure for the evaluation and comparison of water-testing filters intended for the enumeration of specific organisms and mixed microbial populations. The procedure provides general guidelines for comparative testing of the recoveries of bacteria, yeasts and other fungi on membrane filters, as compared to recoveries by the spread plate and pour plate techniques.

This part of ISO 10705 specifies a method for the detection and enumeration of bacteriophages infecting Bacteroides fragilis by incubating the sample with an appropriate host-strain. The method is applicable to all kinds of water, sediments and sludge extracts, where necessary after dilution. In the case of low phage numbers, a preconcentration step may be necessary for which a separate International Standard has been developed. The method is also applicable to shellfish extracts. NOTE It is desirable for International Standards to be adopted as widely as possible. This part of ISO 10705 includes reference to alternative procedures which obviate the need for expensive materials or equipment which may not be readily available in developing countries. Use of these alternatives will not affect the performance of this method.

This part of IS0 10705 specifies a method for the detection and enumeration of F-specific ribonucleic acid (RNA) bacteriophages by incubating the sample with an appropriate host strain. The method can be applied to all kinds of water, sediments and sludges, where necessary after dilution. In the case of low numbers, a preconcentration step may be necessary for which a separate part of IS0 10705 will be developed. The method can also be applied to shellfish extracts. Depending on the relative abundance of Fspecific RNA bacteriophages to background organisms, additional confirmatory tests may be necessary and are also specified in this part of IS0 10705. The presence of F-specific RNA bacteriophages in a water sample generally indicates pollution by wastewater contaminated by human or animal faeces. Their survival in the environment, removal by widely used water treatment processes and concentration or retention by shellfish resembles that of foodborne and waterborne human enteric viruses, for example the enteroviruses, hepatitis A virus and rotaviruses.

This International Standard specifies a method that is applicable for the detection and enumeration of Cryptosporidium oocysts and Giardia cysts in water. It is applicable for the examination of surface and ground waters, treated waters, mineral waters, swimming pool and recreational waters. This method does not allow identification to species level, the host species of origin or the determination of viability or infectivity of any Cryptosporidium oocyst or Giardia cyst which may be present. These procedures are for use by experienced analysts who have successfully completed competency tests prior to commencing analysis. In addition, such analysts should continue to demonstrate competency by examining seeded samples at regular intervals and taking part in external quality assurance schemes. NOTE Bodies resembling Cryptosporidium or Giardia in morphology can be present and these may be mistaken for oocysts or cysts. Results should be interpreted with care. Where there is doubt about the identity of oocysts or cysts or where an unusually high result is obtained, it is advisable to have the slides examined by experts from other laboratories to confirm or refute the findings.

This part of ISO 10705 specifies the general principles for assessing the performance of methods for the concentration of bacteriophages from water. Concentration is recommended for those water samples expected to contain < 3 pfp (plaque-forming particles) per millilitre. Concentration methods can be applied to all kinds of water provided that the amount and nature of suspended solids and/or dissolved matter do not interfere with the concentration procedure. This part of ISO 10705 does not give specific details of concentration methods, but outlines the fundamental principles for evaluating the suitability of a particular method for a given type and volume of water. Annex A gives examples of methods that have been found satisfactory and their fields of application
