动态浊度法定量检测乳糖一水合物中细菌内毒素的含量

摘要 目的：应用美洲鲎试剂（Limulus Amebocyte Lysate，LAL）的动态浊度法，定量检测乳糖一水合物中细菌内毒素。方法：依据
2015 年版《中华人民共和国药典》<1143 细菌内毒素检查法 > 的要求，应用Pyros Kinetix®
Flex（PKF）细菌内毒素定量检测系统，使用葡聚糖抑制缓冲液稀释溶解LAL，建立细菌内毒素的标准曲线并确定稀释倍数，通过测定供试品溶液中外加内毒素的回收率进行干扰试验，并定量测定样品中的细菌内毒素含量。Pyros®
EQS 软件自动绘制并出具标准曲线，计算溶液中内毒素的绝对含量。
结果：内毒素检测标准曲线浓度线性范围为 0.001~0.032 EU·mL-1（r=-0.9854），标准曲线回归方程为 logT=-0.200logC+2.943；供试品在稀释 100、300、500 倍时均对试验无干扰作用，细菌内毒素回收率在50%~200%之间；4 批样品的内毒素定量测定结果符合规定。
结论：在PyrosKinetix®Flex（PKF）细菌内毒素定量检测系统平台，使用 LAL
建立的动态浊度法，可用于乳糖一水合物的细菌内毒素定量检测。LAL 有更高的灵敏度，葡聚糖抑制缓冲液可有效阻止由(1 → 3)-β-D-
葡聚糖引起的干扰，提高试验的成功率，符合数据完整性的要求。

Abstract Objective：To develope a kinetic
turbidimetric method with Limulus Amebocyte Lysate（LAL) to quantify
bacterial endotoxin in lactose monohydrate. Methods：According
to the requirements in Chapter 1143 Bacterial Endotoxin Test in Chinese
Pharmacopoeia 2015 edition，by using Pyros Kinetix® Flex（PKF）bacterial
endotoxin quantitative detection system and LAL dissolved in glucan
inhibitory buffer，a standard curve for bacterial endotoxin was
established and the dilution factor was determined.The interference test
was performed by measuring recovery rate of endotoxin added in the test
solution，and bacterial endotoxin in the sample was quantitatively
determined.Pyros® EQS software automatically produced a calibration
curve to calculate the absolute content of endotoxin in each solution. Results：Concentration linear ranges of endotoxin detection calibration curve was
0.001-0.032 EU·mL-1（r=-0.985 4）with recovery rate of
50%-200%，and no interference was observed for test samples diluted
100-500 times.Results of quantitative determination of endotoxin in four
batches of samples met the requirements.Conclusion：On
the platform of PKF bacterial endotoxin quantitative detection
system，the kinetic turbidimetric endotoxin method using LAL can be used
for bacterial endotoxin detection of lactose monohydrate.
Glucan inhibitory buffer effectively prevented the interference caused by（1 → 3）-β-D-glucan，improved the test success rate，sensitivity and accuracy of detection.the experiment met data integrity requirements.

注射用福沙匹坦二甲葡胺是用于抑制肿瘤化疗所致呕吐反应的药物，以静脉注射的方式给药。由于铂类、阿霉素等多种抗肿瘤药物在使用过程中会产生严重的呕吐反应，而急性剧烈的恶心、呕吐可能导致患者脱水、电解质紊乱、营养不良，严重者可能因消化道黏膜损伤而出血、感染甚至死亡，严重影响化疗效果，因而止吐药是抗肿瘤治疗重要的辅助治疗药物。福沙匹坦二甲葡胺注射剂与其他止吐药联用，可用于防治中等催吐及大剂量催吐的肿瘤化疗（包括大剂量顺铂）初始和反复用药引起的急性和迟后的恶心和呕吐［3］。
乳糖一水合物是一种白色或类白色的晶体状粉末，缓慢溶于水，几乎不溶于乙醇（96%），是注射用福沙匹坦二甲葡胺注射液的一类重要辅料。对乳糖一水合物进行细菌内毒素检测，是对药物生产源头的质量把关与控制，可使药物的质量更有保证，故在福沙匹坦二甲葡胺注射剂生产的质量控制中具有重要意义［2］。目前国内多采用凝胶法对乳糖一水合物进行细菌内毒素检测，质量标准规定其限值应小于0.005 EU·mg-1。本研究参照 2015 年版《中华人民共和国药典》<1143 细菌内毒素检查法 >［1］中的动态浊度法，对乳糖一水合物中的细菌内毒素定量检测方法进行开发，试图找到一种干扰影响小、灵敏度高、更为准确的定量检测方法。
1 仪器与试药
Pyros Kinetix® Flex 96（PKF 96）细菌内毒素定量检测系统（Associates of Cape Cod 公司)。Pyros® EQS 软件（搭配 PFK 细菌内毒素定量检测系统的专用软件，Associates of Cape Cod.Inc.）
乳糖 一 水 合 物（DMV Fonterra Excipients GmbH &Co.KG 公 司，Lactohale® 200 Milled lactose monohydrate，批号：Lactose-1031D73、Lactose-1031D74、Lactose-1031D75、Lactose-1031D76）。
动 态 浊 度 法 美 洲 鲎 试 剂（LAL，批 号 518-11-903-T，灵敏度λ=0.001 EU·mL-1，规格 5 mL·瓶 -1）；细菌内毒素工作标准品（control standard endotoxin，CSE；批号 162）；无热原水（LAL reagent water，LRW批 号 AD17657282，规 格 50 mL· 瓶 -1）；葡 聚 糖 抑制缓冲液（批号 1207063）；硼硅酸盐玻璃反应试管（8 mm×75 mm）；硼硅酸盐玻璃稀释试管（12 mm×
